Q.

Which one of the following statement is not true regarding gel electrophoresis technique?

  1. A

The process of extraction of separated DNA strands from gel is called elution.

  2. B

The separated DNA fragments are stained by using ethidium bromide.

  3. C

The presence of chromogenic substrate gives blue coloured DNA bands on the gel.

  4. D

Bright orange coloured bands of DNA can be observed in the gel when exposed to UV light.

Solution

Gel electrophoresis separates DNA fragments by size in an electric field through an agarose gel.

Bands are visualised by staining with ethidium bromide, which intercalates into the DNA and fluoresces bright orange under UV light. Recovery of a specific band from the gel is called elution.

Chromogenic substrates that give blue products are associated with enzyme-linked detection methods (such as ELISA or western blotting), not with routine DNA gel electrophoresis. Statement (3) is therefore not true.
