Q.

Given below are two statements :

Statement I : The DNA fragments extracted from gel electrophoresis can be used in construction of recombinant DNA.

Statement II : Smaller size DNA fragments are observed near anode while larger fragments are found near the wells in an agarose gel.

In the light of the above statements, chocse the most appropriate answer from the options given below :

  1. A

Statement I is incorrect but statement II is correct

  2. B

Both statement I and statement II are correct

  3. C

Both statement I and statement II are incorrect

  4. D

Statement I is correct but statement II is incorrect

Solution

Statement I: Restriction-digested DNA is run on agarose gel, the desired band is cut out, and the DNA is recovered by elution. These purified fragments are then ligated into cloning vectors recombinant DNA. Statement I is correct.

Statement II: Agarose gel acts as a sieve. DNA is negatively charged and migrates toward the anode (+). Smaller fragments slip through the matrix more easily and travel farther (closer to the anode), while larger fragments lag behind near the wells (cathode end). Statement II is correct.

Both statements are correct.
